Biological Information
Background Information:
YopH is a Yersinia protein that has protein tyrosine phosphate phosphatase activity (PTPase). YopH is required for virulence and has homology to eukaryotic SH2 proteins and SH3 binding domains. YopH breaks down signal transduction mechanisms in immune cells and inhibits the immune response. It contributes to the ability of the bacteria to resist phagocytosis by peritoneal macrophages.

Testing Information
Procedure Summary:
Recombinant protein phosphatase YopH expressed in E. coli cells is used. Test compound and/or vehicle is preincubated with 0.013 μg/ml enzymeõ for 15 minutes at 37°C in modified HEPES buffer pH 7.2. The reaction is initiated by addition of 20 μM DiFMUP for 30 minutes. Determination of the amount of DiFMU formed is read spectrofluorimetrically at 358 nm/450 nm. Compounds are screened at 10 μM.
