Tumor Cell Proliferation Assay in PLC/PRF/5 Human Liver Cells, Panlabs

Procedure Summary:

Human liver hepatoma PLC/PRF/5 cells (ATCC, CRL-8024), (3 x 10^4/ml in growth medium), are pre-incubated in 96-well plates in an atmosphere of 5% CO2 at 37°C overnight. Test compound and/or vehicle is then added for an additional 72 hours. After the 72-hour incubation, CellTiter-Glo reagent is added and for a 10 minute incubation luminescence is measured using a microplate reader. Growth inhibition (GI50), total growth inhibition (TGI) and the 50% lethal concentration (LC50) are reported.

Criteria For Significance:

≤ 50% Cell Growth

Test Concentration / Dose:

Compounds are screened at 30, 10, 3, 1, 0.3, 0.1, 0.03 and 0.01 μM

Test Sample Requirements:

75 μl of 10 mM stock or 1 mg (pre-weighed) powder (assuming FW ≦500 and purity 100%)
