Background Information:

The testis- and skeletal-muscle-specific dual-specificity phosphatase (TMDP), also called DUSP13, is a member of the dual-specificity phosphatase (DUSP) subgroup of protein tyrosine phosphatases which catalyze dephosphorylation of protein substrates at both tyrosine and threonine residues. TMDP is highly expressed during meiosis and is mainly localized in spermatocytes. TMDP is supposed to be involved in the regulation of meiosis and/or differentiation of testicular germ cells during spermatogenesis.

Procedure Summary:

Human recombinant protein phosphatase TMDP expressed in E. coli cells is used. Test compound and/or vehicle is preincubated with 0.054 μg/ml enzymeǂ for 15 minutes at 37°C in modified sodium acetate buffer pH 4.5. The reaction is initiated by addition of 10 μM DiFMUP for 30 minutes. Determination of the amount of DiFMU formed is read spectrofluorimetrically at 358 nm/450 nm. Compounds are screened at 10 μM.
