Biological Information
Background Information:
RPTPg (PTPRG, protein tyrosine phosphatase receptor type G) a member of the protein tyrosine phosphatase (PTP) family. RPTPg have high substrate specificity for phosphotyrosyl proteins, at the primary sequence level sharing little similarity with the protein serine phosphatases, protein threonine phosphatases, or the acid and alkaline phosphatases. PTP gamma has been implicated in playing a tumour suppressor role in kidney, lung and breast cancers. .

Procedure Summary:
Human recombinant protein phosphatase RPTPg expressed in E. coli is used. Test compound and/or vehicle is preincubated with 0.63 ng/ml enzyme‡ for 15 minutes at 37°C in modified Tris-HCl buffer pH 7.2. The reaction is initiated by addition of 100 μM DiFMUP for 60 minutes. Determination of the amount of DiFMU formed is read spectrofluorimetrically at 358 nm/450 nm. Compounds are screened at 10 µM.
