Biological Information
Background Information:
Protein phosphatase 5 (PP5) is a member of the PPP family of serine/threonine phosphatases, consists of a C-terminal catalytic domain and an N-terminal TPR (tetratricopeptide repeat) domain which is involved in protein-protein. PP5 has been implicated in wide ranging cellular processes including MAPK-mediated growth and differentiation, cell cycle arrest and DNA damage repair, regulation of ion channels, the cellular heat shock response and steroid receptor signaling.

Testing Information
Procedure Summary:
Human recombinant protein phosphatase PP5 expressed in E. coli cells is used. Test compound and/or vehicle is preincubated with 3.38 µg/ml enzymeǂ for 15 minutes at 37°C in modified HEPES buffer pH 7.2. The reaction is initiated by addition of 200 μM DiFMUP for 30 minutes. Determination of the amount of DiFMU formed is read spectrofluorimetrically at 358 nm/450 nm. Compounds are screened at 10 μM.
