Background Information:
Protein kinase Cg (PKCg) is one of the major protein kinase C isoforms of a family of serine/threonine protein kinases. Breakdown of phosphatidylinositol leads to formation of diacylglycerol and inositol triphophate, the former acting synergistically with Ca2+ to activate protein kinase C (PKC), resulting in translocation of the enzyme from cytosol to the membrane. Phosphorylation of proteins by PKC has been implicated as a pivotal regulatory element in signal transduction, cellular regulation, and with a high affinity to phorbol ester tumor promoters.

Procedure Summary:
Human recombinant protein kinase PKCg expressed in Sf21 insect cells is used. Test compound and/or vehicle is preincubated with 0.474 μg/ml enzyme‡ in modified HEPES buffer pH 7.4 for 15 minutes at 37⁰C. The reaction is initiated by addition of 250 μg/ml histone H1 , 10 μM ATP and 0.25 μCi [γ32P]ATP for another 30 minute incubation period and terminated by further addition of 3% H3PO4. An aliquot is removed and counted to determine the amount of [32P]Histone H1 formed. Compounds are screened at 10 μM.
