Biological Information

Background Information:

Mitogen-activated protein (MAP) kinases are proline-directed protein kinases that mediate the effects of numerous extracellular stimuli on a wide array of biological processes, such as cellular proliferation, differentiation, and death. Three groups of mammalian MAP kinases have been studied in detail: the extracellular signal-regulated kinases (ERK), the c-Jun NH2-terminal kinases (JNK), and the p38 MAP kinases. The p38 MAP kinase group, which includes the isoforms p38a, p38b, p38g and p38d, activates the transcription factors CREB, ATF1, ATF2, CHOP, and MEF-2C. The p38 MAP kinase also activates other protein kinases, such as Mapkap-2, Mapkap-3, and Mnk1/2.

Testing Information

Procedure Summary:

SAPK2b (h) is incubated with 25 mM Tris pH 7.5, 0.02 mM EGTA, 0.33 mg/mL myelin basic protein, 10 mM MgAcetate and [gamma-33P]-ATP (specific activity and concentration as required). The reaction is initiated by the addition of the Mg/ATP mix. After incubation for 40 minutes at room temperature, the reaction is stopped by the addition of phosphoric acid to a concentration of 0.5%. An aliquot of the reaction is then spotted onto a filter and washed four times for 4 minutes in 0.425% phosphoric acid and once in methanol prior to drying and scintillation counting.
