Background Information:

Neurotoxicity refers to the damage and disruption of nervous system function or structure caused by exogenous compounds, potentially leading to acute or chronic toxic effects, particularly during critical developmental stages. The SH-SY5Y human neuroblastoma cell line has been widely utilized as an in vitro model for neurotoxicity studies due to its adaptability. It can be maintained as proliferative neuroblasts or induced to differentiate into neuron-like cells, mimicking aspects of neuronal function and morphology. This differentiation capability enables assessment of neurotoxicity in both immature and mature neuronal states. As a human-derived neuronal-like model, differentiated SH-SY5Y cells provide a physiologically relevant platform for neurotoxicological screening, supporting the evaluation of drug safety, chemical risk assessment, and mitigation of adverse effects on the nervous system. In this assay, differentiated SH-SY5Y cells are used, and neurite length is assessed to indicate the potential neurotoxicity of the test samples.

Procedure Summary:

SH-SY5Y cells are seeded in the 96-well half area plate and differentiated for 7 days at 37°C, 5% CO₂. Test compound and/or vehicle is then added for an additional 72 hours at 37°C, 5% CO₂. After incubation, the cells are stained with fluorescent dyes. The cell images are acquired by ImageXpress Micro Confocal High-Content Imaging System. Neurite outgrowth is evaluated and analyzed by MetaXpress software. Decrease of neurite outgrowth by 50 percent or more (≥50%) relative to vehicle treated control indicates significant neurotoxicity. 1 μM Brefeldin A is defined as 100% inhibition.
