Biological Information
Background Information:
Lambda protein phosphatase is a Mn2+-dependent protein phosphatase with activity towards phosphorylated serine, threonine and tyrosine residues. The bacteriophage lambda enzyme exhibited a high degree of similarity to the catalytic subunits of a variety of protein serine/threonine phosphatases, many of which have recently been shown to play critical roles in diverse cellular processes such as growth and differentiation, oncogenesis, DNA replication, transcription, and protein synthesis.

Testing Information
Procedure Summary:
Recombinant protein phosphatase lPP expressed in E. coli cells is used. Test compound and/or vehicle is preincubated with 0.002 μg/ml enzymeǂ for 15 minutes at 37°C in modified HEPES buffer pH 7.2. The reaction is initiated by addition of 10 μM DiFMUP for 30 minutes. Determination of the amount of DiFMU formed is read spectrofluorimetrically at 358 nm/450 nm. Compounds are screened at 10 μM.
