Biological Information
Background Information:
Hematopoietic protein tyrosine phosphatase (HeTP) protein, also called protein tyrosine phosphatase non-receptor type 7, PTPN7, LC-PTP, LPTP, is a cytosolic enzyme expressed strongly in T cells. HePTP plays a negative role signaling by down-regulating MAPK activity and responsible for many different types of signaling including reaction to stress and inflammation and cell migration. Amplification and overexpression of HePTP may be an important cofactor contributing to abnormal myeloid cell growth.

Testing Information
Procedure Summary:
Human recombinant protein phosphatase HePTP expressed in E. coli cells is used. Test compound and/or vehicle is preincubated with 0.016 μg/ml enzymeõ for 15 minutes at 37°C in modified HEPES buffer pH 7.2. The reaction is initiated by addition of 10 μM DiFMUP for 30 minutes. Determination of the amount of DiFMU formed is read spectrofluorimetrically at 358 nm/450 nm. Compounds are screened at 10 μM.
