Biological Information
Background Information:
MAPK phosphatases (MKPs) are a group of dual specific phosphatases (DUSPs) which deactivate the mitogen-activated protein kinases (MAPKs) via dephosphorylation of phosphotyrosine and phosphothreonine residues. Dual specificity phosphatase 22, (DUSP22), also called JKAP, JSP1 and MKPX is a part of the protein-tyrosine phosphatase family, plays a role in various inflammatory and have been shown to be important in regulating immune responses.

Testing Information
Procedure Summary:
Human recombinant protein phosphatase DUSP22 expressed in E. coli cells is used. Test compound and/or vehicle is preincubated with 2 μg/ml enzymeǂ for 15 minutes at 37°C in modified HEPES buffer pH 7.2. The reaction is initiated by addition of 10 μM DiFMUP for 30 minutes. Determination of the amount of DiFMU formed is read spectrofluorimetrically at 358 nm/450 nm. Compounds are screened at 10 μM.
