Biological Information
Background Information:
RAS is a commonly mutated oncogene family in cancers. Among three RAS isoforms (KRAS, NRAS, and HRAS), approximately 85% of mutations contribute from KRAS. Codon 12 (G12) is the most frequent mutation in KRAS. Notably, G12D is the most prevalent, followed by G12V and G12C. Depending on cancer types, pancreatic (~90%), lung (~85%), and colorectal (~68%) cancers are highly driven by KRAS G12 mutations, while melanoma (~85%) is highly associated with NRAS Q61 mutations. With the recent breakthrough of turning undruggable KRAS G12C into a druggable target, other RAS variants also possess high therapeutic potentials for treating various cancers. This panel includes a total of 14 NEA-based assays, which cover most KRAS variants, including G12D, G12V, G12C, G12R, G12A, G12S, G13D, G13C, and Q61H, in addition to NRAS Q61R and HRAS G12V variants. By screening this panel, potency, and selectivity across RAS variants can be efficiently evaluated and identified.
