Background Information:
The L-type voltage-gated calcium channel is assayed on the FDA-preferred CiPA Protocol. Entry of extracellular Ca2+ through L-type Ca2+ channels is the trigger for cardiac muscle contraction via Ca2+-induced Ca2+ release from the sarcoplasmic reticulum. Concomitant block of other cardiac ion channels such as the L-type Ca2+ channel may reduce one of the substrates for Torsades de Pointes, increased QT dispersion (Antzelevitch, 2005a) that would result from hERG block alone (Antzelevitch, 2005a; Antzelevitch, 2004; Drouin et al., 1998). Furthermore concomitant block of L-type channels may suppress the essential trigger for Torsades de Pointes, early afterdepolarizations, caused by reactivation of Ca2+ current during phase 2/3 of the cardiac action potential (January and Riddle, 1989).
