15-Lipoxygenase initiates the metabolic pathway leading to the formation of lipoxins by converting arachidonic acid to 15-hydroxyperoxy-eicosatetraenoic acid (15-HPETE). Lipoxins may share some of the bioactivity associated with leukotrienes.

Procedure Summary:
Human recombinant 15-LO expressed in E. coli cells is used. Test compound or vehicle are combined with 16 µg/mlǂ of enzyme for 15 minutes at 37°C in Tris buffer pH 7.4. The reaction is initiated by addition of 10 µM arachidonyl amide and dye DHR123 for 30 minutes at 37°C. Read plate on a spectrophotometer (Excitation: 485 nm, Emission: 535 nm). Compounds are screened at 10 µM.
