Biological Information
Background Information:
Protein tyrosine phosphatase, non-receptor type 22, also known as PTPN22, LYP, is a protein that in humans is encoded by the PTPN22 gene. PTPN22 affects the responsiveness of T and B cell receptors, and mutations are associated with increases or decreases in risks of autoimmune diseases.

Testing Information
Procedure Summary:
Human recombinant protein phosphatase PTPN22 expressed in E. coli cells is used. Test compound and/or vehicle is preincubated with 0.017 μg/ml enzymeǂ for 15 minutes at 37°C in modified HEPES buffer pH 7.2. The reaction is initiated by addition of 10 μM DiFMUP for 30 minutes. Determination of the amount of DiFMU formed is read spectrofluorimetrically at 358 nm/450 nm. Compounds are screened at 10 μM.
