Biological Information
Background Information:
A free radical is an atom or molecule that has one or more unpaired electrons. The evidence that reactive species of oxygen (O2-, H2O2, OH- and NO2+) play an important role in pulmonary oxygen toxicity, inflammation, neurodegenerative disorders, ischemia/reperfusion injury and cancer, etc. The chromogen 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulphonic acid) radical (ABTS+) can be directly generated by the enzymatic system formed by hydrogen peroxide and horseradish peroxidase in ethanolic media. The ABTS+ generated shows excellent stability for at least 10 minutes, which was used for estimating anti-oxidant activity. This screen can provide an independent and simultaneous evaluation of the low molecular weight and protein antioxidants present in biological samples such as serum.

Testing Information
Procedure Summary:
Test compound and/or vehicle is incubated with ABTS+ solution (ABTS substrate buffer : H2O2 : Peroxidase = 1 : 1 : 1) for 5 minutes at 25⁰C. Scavenger activity is determined spectrophotometrically by measuring the quenching of the chromagoen radical ( ABTS+) . Compounds are screened at 10 μM.
