Background Information:

Diamine oxidase (DAO, also known as amiloride binding protein (ABP1), amine oxidase copper containing-1 [AOC1]) is a member of oxidases family, an important regulator of polyamine levels. It is found in various tissues, especially active in the intestinal mucosa. DAO deficiency or inhibition can result in histaminosis or dietary histamine intolerance and the associated symptoms.

Procedure Summary:

Human recombinant DAO enzyme expressed in Sf21 cells is used. Test compound and/or vehicle is preincubated with 500 ng/ml active enzymeó in modified HEPES buffer pH 7.5 for 15 minutes at 25°C. The reaction is initiated by addition of 5 μM histamine substrate for another 30 minute incubation period. Determination of the amount of resorufin formed is read spectrofluorimetrically at 544 nm/590 nm. Compounds are screened at 10 μM.
