Biological Information

Background Information:

STK35 (Serine/threonine kinase 35) is a protein kinase that catalyzes the phosphorylation of serine and threonine residues on target proteins, thereby modulating their activity and function. This kinase interacts with nuclear proteins, playing a significant role in chromatin remodeling and transcriptional regulation. STK35 has been implicated in cellular stress responses and DNA damage repair pathways, suggesting its involvement in maintaining genomic stability. Overexpression of STK35 has been observed in colorectal and breast cancers, correlating with poor prognosis, which indicates its potential as a prognostic biomarker. The multifaceted functions of STK35 in cell cycle regulation, apoptosis, and signal transduction pathways make it an attractive therapeutic target for cancer and other diseases characterized by dysregulated cell signaling. However, inhibition of STK35 may be associated with cardiotoxicity, including hypertension, QT prolongation, and arterial thrombosis.

Testing Information

Procedure Summary:

Human recombinant protein kinase STK35 expressed in SF21 insect cells is used. Test compound and/or vehicle is preincubated with 2.94 μg/ml enzymeǂ in modified MOPS buffer pH 7.2 for 15 minutes at 37oC. The reaction is initiated by addition of 200 nM RS repeat peptide, 10 mM ATP and 0.05 mCi [g32P]ATP for another 30 minute incubation period and terminated by further addition of 3% H3PO4. An aliquot is removed and counted to determine the amount of [32P]RS repeat peptide formed. Compounds are screened at 10 mM.
