Biological Information
Background Information:
Voltage-gated calcium channels mediate calcium influx in response to membrane depolarization and regulate intracellular processes such as contraction, secretion, neurotransmission, and gene expression. They are members of a gene superfamily of transmembrane ion channel proteins that includes voltage-gated potassium and sodium channels. Pharmacology of the three families of calcium channels is quite distinct. N-Type calcium channels are relatively insensitive to L-Type channel blockers and are specifically blocked with high affinity by peptide toxins from spiders and marine snails. The peptide toxins are potent blockers of synaptic transmission.

Testing Information
Procedure Summary:
This assay measures binding of [¹²⁵I]Conotoxin GVIA to calcium channel N-Type receptors. Frontal lobe brain membranes of male Wistar derived rats weighing 175 ± 25 g are prepared in modified Tris-HCl pH 7.4 buffer using standard techniques. A 40 μg‡ aliquot of membrane is incubated with 10 pM [¹²⁵I]Conotoxin GVIA for 30 minutes at 4℃. Non-specific binding is estimated in the presence of 100 nM w-conotoxin GVIA. Membranes are filtered and washed 3 times and the filters are counted to determine [¹²⁵I]Conotoxin GVIA specifically bound. Compounds are screened at 10 µM.
Note: ‡Membrane protein may change from lot to lot, the concentration used will be adjusted if necessary.
