Biological Information
Background Information:
Caspase 2 is a member of the CED-3/caspase-3 subfamily. Cleavage sites for caspase 2 are located at asp316, asp330, and possibly asp152. Caspase 2 appears to be localized in the Golgi as well as the nucleus, and is released when the membranes disintegrate, early in the apoptotic process. Caspase inhibitors are currently sought as therapeutics for diseases such as Alzheimer's, Parkinson's and cerebral/myocardial ischemia.

Testing Information
Procedure Summary:
Human recombinant caspase 2 expressed in E. coli is used. Test compound and/or vehicle is preincubated with 0.83 U/ml enzyme‡ in modified HEPES buffer pH 7.4 for 15 minutes at 37⁰C. The reaction is initiated by addition of 25 μM Z-VDVAD-AFC for another 60 minute incubation period. Determination of the amount of AFC formed is read spectrofluorimetrically at 400 nm/510 nm. Compounds are screened at 10 μM.
