Background Information:
Knowledge of the partitioning behavior of a therapeutic compound into red blood cells is important to the interpretation and understanding of the compound’s pharmacokinetic (PK) profile. A high partitioning ratio may also indicate accumulation of the compound in red blood cells and potential hematotoxicity. The RBC-to-plasma ratio (KRBC/PL) is obtained by incubation of the test compound in whole blood and determination of the amount of compound in plasma and reference plasma via LCMS analysis.
